Active Ingredients: Glibenclamide(glyburide)

Therapeutic actions: Glyburide oral inhibits ATP-sensitive potassium channels which are located in the pancreatic beta cells. The following process causes cell membrane depolarization, which opens the voltage-dependent calcium channels, thus triggering an increase in intracellular calcium which stimulates insulin release.

Indications: Glyburide is used for Insulin-dependent Diabetes type 2 with partially preserved synthesis of insulin.

Contraindications and cautions: Glyburide should be used with caution in the following cases:

  • Insulin-dependent diabetes;
  • diabetic ketoacidosis;
  • pregnancy;
  • patients with severe renal and liver dysfunction;
  • registered individual hypersensitivity to Glyburide.

Elderly patients with hepatic and renal failure should take Glyburide with care, in order to prevent hypoglycemia. During the treatment no alcohol should be consumed; otherwise the risk of hypoglycemia exists. Glyburide is excreted in mother milk, there for breastfeeding should be discontinued.

Adverse effects:
There are possible side-effects associated with this medicine that can affect individuals in different ways. If a side effect is stated here, that does not necessarily mean the fact that all people using Glyburide will experience it or any other.
Stop using glyburide and seek medical attention if an allergic reaction occurs (breathing difficulty; closing of the throat; lip, tongue, or face swelling; or hives).

Other side effects from glyburide result in changed blood sugar levels that are either too high or too low. Low blood sugar occurs when too much glyburide is taken(meals are missed or delayed); if other medications are taken; during illness, especially with vomiting or diarrhea; after drinking alcohol;if you exercise more than usual; and in other situations.
Increased blood sugar may occur when not enough glyburide is taken; if you exercise less than usual; during fever or other illness;if you take other medications; if you eat significantly more food than usual; and in other situations.

In concomitant administration of β-blockers, H2-blockers, sulphonamides, cumarin derivatives, tetracycline, clofibrate, chloramphenicol, salicylates, and MAO-inhibitors the hypoglycemic effect of the Glibenclamid is increased. Glibenclamid potentiates anticoagulant effect of warfarin. Corticosteroids, estrogen and gestegen hormones, phenothiazines, and oral contraceptives cause a decrease of the Glibenclamid effects.
